Challenge Cup 20/21

we know that Spain, Italy won't be involved; Bucharest Wolves denied too

to me it looks like Germany and Portugal are EPCR's only options now

It's a mystery. Frankfurt 1880 were looking for an international competition so they could be one of the teams. Maybe a Georgia team?

If Italy is excluded due to the general uncertain situation it would be surprising if EPCR is going to invite teams from T2 nations.
Anyway, the man who said (to a newspaper) that EPCR sent him a mail with the decision to exclude this year the italian Top12 clubs was the president of the Italian Federation itself, so it's hard to have doubts about his words

Just 14 teams in the Challenge Cup 2020/21: no italian Top12, spanish, romanian or other T2 clubs.
One only round robin with just four games per team and then a round of 16 with the eight Champions Cup teams not qualified to the quarterfinals in their competition; then quarterfinals, semifinals, final

According to the netherlander technical director, Rugby Europe is going to inaugurate a new competition for T2 clubs, involving one club for every nation of the RE Championship and one club from each of the two best nations of the RE Trophy

Netherlands, says the t.d., is mumbling to field a sort of national team, like the Sunwolves, rather than a club.

This could mean that the italian Top12 clubs will be trapped out of Europe, if EPCR confirmed also in the next seasons that there's no place for them in Challenge Cup

Any reason to think this new tournament would have a relationship to challenge cup?

No, not explicitly, it was just the first thread vaguely related that I found.
Anyway according to the first posts of the thread there could be still place for two not Premiership/Top14/Pro14 teams in Challenge, and it seems likely that this new competition could work as qualifier
The qualified teams could come both from this competition or one from this competition and one from the Italian championship. But these are just hypothesis
